When I create new file in $HOME/Desktop directory, the desktop icon is not created automatically - I have to press F5 to see new file.

I'm using Xubuntu 6.06.1, xfce installed using xfce graphical installer. Previously I used xfce 4.3.90.1 without such problem.

Be sure gamin is installed, and that thunar-vfs is compiled with gamin support.  FYI, this is a bug report for ubuntu, not for xfce.  Go report it there.
